i think the only way to sort it out would be to take a look inside .........don`t think that was the answer you wanted to hear ...........but a knocking could be a thousand and one things
never squat down when wearing spurs !!
can you make the noise so we can have a better idea...e.g. crash /bang/wallop.

It sounds like If you were holding a layshaft in you hands complete with gears and selector, and you twist the layshaft back and fourth,The knock/donk sound you would get from the gears/gear hitting the seletor?
not sure if that makes any sense to anyone?

Anytime you have a 50-50 chance of getting something right, there`s a 90% probability you`ll get it wrong.
so it"s a donk eh.

might be time to stop listening and start looking inside; i heard a crash bang wallop noise in my motor which rapidly tuned into a whack snap crunching sound followed by a imediate sort of OOuchhh noise as i skided to a halt; want anything serious only crank barrel piston head pot flywheel stator and crankcase needed replacing when the rod snapped and went out through the crankase demolishing everything except the gearbox and sprockets,
keep listening; muppet,
keep listening; muppet,
C’est la vie

is there any gear oil in the crank case??
If not could be the gears meshing dry.
Or possibly the kick start ratchet just catching on 1st gear
