As already said, do a leak test but I'd also check the crank especially the big end bearing.

No mention of the carb so far Bilko :? Has the same one been used on both engines? ie, just pulled off the rubber and then slotted onto the next engine? I had a lot of 'fun' as well earlier this year trying to start my Avanti after a rebuild, so much so that I managed to bust my kickstart in the process  Changed everything like you but eventually found the problem to be that the nipple had become disengaged for the choke, must have happened whistle the top of the carb had been removed and left hanging with the cables still connected  Your problem could be float related, have you checked for good fuel flow through the carb i.e. float bowl removed and a bowl under to catch the fuel as it runs through? check for a leaky or sticking float as well.

Yes carbs changed Colin. 3 different ones.

Cranks going to come out but haven't got anything decent enough to replace it apart from a 60mm one.

Carb changed but did the top and choke mechanism stay the same? Logic suggests that the problem is on the bike not he engine

Shirlock Holmes said "when you have eliminated the obvious, whatever is left, no matter how unlikely, must be the answer".......I think you have the obvious covered so you need to start looking at the unlikely now ;) ....Check your tyre pressures next
