Do diaphragm carburettors only work with a reed valve
or will they work with just a pulse from the crank case no reed valve?

Just looking at weather one of these could cure clearance issues.

By diaphragm carburettor, do you mean one that does away with a float bowl and replaces it with a flow of fuel delivered by a diaphragm pump which is operated by pressure pulses within the crankcase?

Such carbs are common on jet-skis.

And Karts

Used to race, we used Tilliston carbs in 100cc Briton class, they just bolted on to rotary valve cases.
Pride has succsefully run these in the past.

Those are the ones.

Did you run a reed valve on the motor or did it run fine
with a vacuum hose from the crank case?

That looks just the Job.
Are you engaged in making it run of a lambretta motor?

I've acquired a crappy Indian 150 engine case and once I've built my TT3
I fancy playing around with one of these carbs.
