Two rear punctures within an hour

Hi all,
I went out on my PX 180 EFL today and managed to puncture the rear tyre. I changed it and then drove for about 2 miles and the same thing happened again. With no obvious puncture marks on the tread of the tyre, I have pulled out the inner tube and it has a large hole on the middle of the inside (engine side) wall of the tube. I have checked for any obvious contact with the main engine block but cannot see where it would foul on the tyre. it sits pretty close to the rear suspension and the clutch housing. Also, I'm running a revolver exhaust with it but once again I cannot see where there is or has been contact.
Any thoughts or advice as to what caused the two punctures and how I can resolve it would be appreciated.
